I have tried to configure remote desktop to use a different port rather than the standard 3389,but nothing is working. I have changed it in the registry and on the router no luck.

I am already using a computer on the standard port and I want to setup another computer on a separate port.

    how do I do ?

Why do you want to change the port number? You can have more then one connection at a time both using 3389.

I am remoting into a network that has 1 IP. From there the port forwarding is set to use port 3389 to one of the designated PCs. If i set both the PCs up on port 3389 theirs no way to determine which computer your going to remote into.

And did you also change the port on the computer you are connecting from?
