I need to load XP on to my wife's business computer to allow some software things to work. Can it be loaded over 98SE?

Yes. If 98 is running smoothly it should give an XP load that runs smoothly.

If you have the option, i would reccomend backing up any important data prior to installing over 98SE since with any install over a previous operating system, there exists an increased chance of painful failure. I don't mean to scare you but it would be best to be prepared for the possibility of needing to do a clean install of XP.
